Author:米倉 真一; Abstract:動物が環境刺激に適切に応答するためには,複雑であるが整然とした神経回路網の存在が必要不可欠である。この神経回路網の形成は,発生過程において,正しい相手と結合することにより形成されるが,その仕組みは分かっておらず,神経科学の学問上でも大きな謎となっている。ショウジョウバエの視覚系は,遺伝学的手法が豊富であり,シナプスの結合に特異性のあることもよく知られていることから,神経回路形成の分子機構解明に際し広く活用されている。近年,ショウジョウバエの行動解析を用いた大規模スクリーニングより,神経回路形成に関与する分子が同定されてきている。本総説では,特異的シナプス形成の分子メカニズムに関する最新の知見について述べると共に,特に,著者が明らかにした特異的シナプス形成におけるNカドヘリンの機能について紹介する。An animal relies on the complex and stereotyped interneuronal connections in its brain to properlys respond to environmental stimuli. Neurons form exquisitely precise patterns of synaptic connections. Hows this is achieved during development remains a central issue in neurobiology. The Drosophila visual systems has been widely used to study the molecular mechanism of neuronal connectivity due to the powerfuls genetic tools that are available in this system. Using visual behavioral screens in Drosophila, recent studiess have identified some of the gene regulatory and cytoskeletal signaling pathways responsible for the layers -specific targeting of Drosophila photoreceptor axons. In this review, I discuss recent progress towardss understanding the molecular mechanisms that control the projection of photoreceptor axons to theirs appropriate target layers in the optic lobe. Especially, I introduce our studies about N-cadherin functionss in the neuronal connectivity.
